Entre las grandes novedades que nos trajo esta entrega, se encuentran: Toadette y Caco Gazapo, con sus propias y peculiares habilidades de juego. Toadette con la posibilidad de transformarse en Peachette gracias a la Super Corona, lo que le da la habilidad de manejarse de mejor manera por el aire y por tierra incluyendo el hecho de contar con más tiempo en el reloj.

Por otra parte, tenemos a Caco Gazapo, un personaje muy divertido con le que también tendremos un extra de tiempo a diferencia de los demás personajes del juego. Una de las cosas interesantes del videojuego es que podremos cambiar de protagonista siempre y cuando estemos fuera de una partida, lo que genera variedad para desplazarnos en los diferentes mundos y optar por cambiar al personaje más conveniente que consideremos.

Por último, y otra de las novedades que nos ofrece Super Mario Bros. U Deluxe es el detalle de vibración en los controles de juego, algo divertido pues nos integra más al juego y nos hace sentir más palpable la experiencia de juego y lo que ocurre dentro de él.
